I would Start with The Habit and then The Lord of the Rings; these stories give one a good introduction to the Tolkien universe and inriches his other writings.
The Lord of the Rings if you don't have a taste for something written as a children's book. Otherwise, start with The Hobbit. Everything in The Hobbit that you need to know to understand what is going on in LotR is summarized in the latter.
I would also start with The Hobbit, since that sets up the Lord of the Rings.
I started with The Silmarillion, but that may be a bit heavy for some people. Unfinished Tales is a book that deserves a lot more love; you may want to give that one a try.
